Specimens for Cytogenetics

 

To ensure a successful outcome please follow the guidelines listed below when collecting specimens. Before shipping, write the patient's name on the container the specimen is to be transported in. When completing the Genetics lab requisition form, please include a reason for study. If FISH studies are desired specify which probe the physician is requesting.

Peripheral Blood

For patients over 6 months of age please collect 3-5 ml whole blood in a sodium heparin tube. This amount is sufficient for karyotyping and FISH studies. For patients under 6 months of age collect 2 ml whole blood in a sodium heparin tube. This amount is sufficient for karyotyping. If the doctor is requesting FISH studies please provide an additional 1 ml of whole blood. Specimen must be kept at room temperature or cooler, do not freeze.

Leukemic Blood

Collect 3-5 ml whole blood in a sodium heparin tube. Please record the most recent white blood cell count on the requisition form. Specimen must be kept at room temperature or cooler, do not freeze.

Bone Marrow

Collect 3-5 ml of bone marrow aspirate in a heparinized syringe. For transport the specimen may be transferred to a sodium heparin tube, a tube with no additives, or a container with RPMI media. Please record the most recent white blood cell count on the requisition form. Fresh specimens must be kept at room temperature, do not freeze.

Amniotic Fluid (Do Not Transport Specimens In Syringes)

Collect 15-20 ml of amniotic fluid for karyotyping. If the doctor is requesting FISH studies include an additional 3 ml of fluid. Collect 5 ml of amniotic fluid for cystic fibrosis screening as well as maternal buccal swabs. Sterilely transfer the fluid to a screw top centrifuge tube to ensure limited exposure of the fluid to the syringes used for collection. Please record the patient's gestational age on the requisition form. Keep the specimen cool but do not freeze.

Skin Biopsy

Provide a full tissue thickness biopsy, approximately 3 mm x 3 mm in size. Collect the specimen in an aseptic manner. The specimen can be transported in: Hanks Balanced Salt Solution, RPMI media, or sterile saline. Please do not use formalin. Keep the specimen cool but do not freeze.

Placenta, Fetal Tissue, or Products of Conception

Collect a 2 cm x 2 cm piece of tissue, it is unnecessary to send more than this amount. The specimen can be transported in: Hanks Balanced Salt Solution, RPMI media, or sterile saline. Please do not use formalin. Keep the specimen cool but do not freeze.

Solid Tumor

The tumor can be tranported in: Hanks Balanced Salt Solution, RPMI media, or sterile saline. Please do not use formalin. Specify the site of origin of the tumor and the type suspected. Keep the specimen cool but do not freeze.

Buccal Swab (FISH testing only and for certain syndromes only, please call before obtaining specimen)

Collect 2 DNA samples with nylon brushes by rubbing the brush firmly along the inside of the cheek for 10-15 seconds. Then place the cap on the brush making sure it is tightly sealed. Keep sample at room temperature. Send the specimen to the genetics lab as soon as possible.
